# Isoxsuprine Hydrochloride
## Overview
Isoxsuprine hydrochloride is a peripheral vasodilator. Its mechanism of action is thought to involve direct relaxation of vascular smooth muscle.
## Primary Indications
* Treatment of peripheral vascular diseases, such as intermittent claudication, arteriosclerosis obliterans, and Raynaud's disease.
* Off-label use in threatened premature labor (tocolysis) has been reported, but is generally not recommended due to lack of efficacy and potential risks compared to established agents.
## Adult Dosing
* **Peripheral Vascular Disease:** 10 mg to 20 mg orally 3 to 4 times daily.
## Pediatric Dosing
* No established pediatric dosing. Use is generally not recommended.
## Dose Adjustments
* No specific dose adjustments are typically required for renal or hepatic impairment, but caution is advised, especially in severe disease.
## Contraindications
* Hypersensitivity to isoxsuprine hydrochloride.
* Should not be administered immediately postpartum or in the presence of fetal distress due to potential risk of maternal hypotension.
## Adverse Effects
* **Common:** Nausea, vomiting, epigastric distress, dizziness, weakness, flushing, hypotension.
* **Less Common:** Tachycardia, palpitations, rash.

* Efficacy in peripheral vascular disease is controversial, and its use has declined significantly due to limited evidence of benefit and the availability of more effective treatments.
* When used for tocolysis, it has largely been replaced by agents like beta-agonists (e.g., terbutaline) and calcium channel blockers, which have shown greater efficacy and a more favorable safety profile.
